    <title>Abstract</title>Muscle atrophy occurs in a variety of physiological and pathological conditions. Specific molecular networks that govern protein synthesis and degradation play important roles in controlling muscle mass under diverse catabolic states. MicroRNAs (miRNAs) were previously found to be regulators of protein synthesis and degradation, and their expressions in skeletal muscle were altered in muscle wasting conditions. However, functional roles of miRNAs in muscle atrophy are poorly understood. In this study, we generated tamoxifen-inducible <italic>Dicer</italic> knockout (iDicer KO) mice and subjected them to 2 weeks of single hindlimb denervation. The expression of <italic>Dicer</italic> mRNA was significantly reduced in muscle of the iDicer KO mice compared to that of WT mice. The loss of <italic>Dicer</italic> moderately reduced levels of muscle-enriched miRNAs, miR-1, miR-133a and miR-206 in both innervated and denervated muscles of the iDicer KO mice. We also found that the extent of denervation-induced muscle atrophy as well as changes of signaling molecules related to protein synthesis/degradation pathways in the iDicer KO mice were comparable to these in WT mice. Taken together, <italic>Dicer</italic> knockout in adult skeletal muscle did not affect denervation-induced muscle atrophy.

  • Beneficial Effects of Yoga Stretching on Salivary Stress Hormones and Parasympathetic Nerve Activity.

    This study aimed to determine the effect of yoga stretching on salivary stress hormones and cardiac autonomic nervous system. To our knowledge, this study is the first to investigate changes in cardiac autonomic nervous system after yoga stretching. In this crossover design study, 10 adult men (age, 26.3 ± 2.5 years) without yoga experience participated in the rest and yoga trials for 90 min. Measurements were carried out before (pre), immediately (post), 60 min, and 120 min after rest or yoga stretching. Saliva samples were collected by chewing a sterile cotton ball at a frequency of 60 cycles per minute. Salivary cortisol and testosterone concentrations were measured using an enzyme-linked immunosorbent assay. With the subjects in the sitting position, heart rate variability was measured using pulse analyzer plus for 150 seconds. As regards rate changes, salivary testosterone level tended to increase (p = 0.088), testosterone/cortisol ratio significantly increased (p < 0.05), and cortisol level significantly decreased (p < 0.05) at 120 min after yoga stretching. The square root of the mean-squared differences of successive normal-to-normal intervals and natural logarithm high-frequency component, which are indicators of parasympathetic nerve activity, increased at 60 min (p < 0.05) and 120 min (p < 0.05) in the yoga trial, respectively. In conclusion, yoga stretching can enhance parasympathetic nerve activity and improve stress hormones. Therefore, yoga stretching may be useful to compensate for physical inactivity and increase life expectancy in the general population.

    Antimicrobial peptides (AMPs) play an important role in innate immunity in human skin. It is known that AMPs mainly function in the stratum corneum. Therefore, AMP concentrations in the stratum corneum need to be precisely measured to clarify functional and physiological importance of AMPs in cutaneous defence. Tape stripping (TS) is a well-established method by which components in the stratum corneum can be collected. However, the usefulness of the TS method for measuring AMP concentration in human skin remains unclear. Therefore, we compared it with another popular method, skin rinsing, which had been established as a method for measuring AMP concentration in human skin. When investigated on healthy medial forearm using RNase 7, which is one of the typical AMPs, as an index, there was a significant positive correlation between RNase 7 concentrations measured by the TS method at adjacent forearm sites, demonstrating the reproducibility of the TS method. Next, a significant positive correlation was detected in RNase 7 concentrations measured using the TS and the skin rinsing method, indicating that the TS method is comparable to the skin rinsing method. Thus, we speculate that the TS method is useful for measuring AMP concentration in human skin.

  • エッセンシャルオイルによる香り刺激が口腔内免疫能に及ぼす影響についての生理心理学的研究

    本研究では、エッセンシャルオイルを用いた香り刺激が安静時の口腔内免疫能および心理状態に及ぼす影響を明らかにすることを目的とした。健康な成人男性16名を対象として、30分間の香り刺激の前後および30分後に口腔内免疫能、自律神経活動、心理ストレス指標を測定した。ベルガモット、ラベンダー、レモンの3種類のエッセンシャルオイルを用いて、噴霧量の強弱に分けた6試行とコントロール試行の合計7試行をランダム化クロスオーバーデザインにて実施した。その結果、ベルガモット「弱」試行にて、香り刺激の30分後に唾液中の分泌型免疫グロブリンA(secretory immunoglobulin A;SIgA)分泌速度の有意な増加がみられた。コルチゾール濃度については、ベルガモット「弱」試行、ベルガモット「強」試行、レモン「弱」試行において有意に低下することが確認された。また、いずれの試行においても心理状態は有意な改善を示し、特にレモン精油の香り刺激は総合的な気分状態を改善することが明らかとなった。したがって、ベルガモット精油は免疫機能とストレスホルモンに有益な効果を示し、総合的な気分状態の改善にはレモン精油が有効であることが示唆された。(著者抄録)

    Protection against age-related immune suppression is important in elderly individuals. This study determined the effect of yoga on mucosal immune function and mental stress. Saliva samples were collected from 23 adult women (age: 60.4 ± 10.4 years) before and after 90 minutes of yoga stretching or rest to measure secretory immunoglobulin A (SIgA), cortisol, and testosterone.   • Changes in Stress and Appetite Responses in Male Power-Trained Athletes during Intensive Training Camp

    An intensive consecutive high-volume training camp may induce appetite loss in athletes. Therefore, this study aimed to investigate the changes in stress and appetite responses in male power-trained athletes during an intensive training camp. The measurements at Day 2 and at the end of a 9-day intensive training camp (Camp1 and Camp2, respectively) were compared with those of the resting period (Rest) and the regular training period (Regular; n = 13). The stress state was assessed based on plasma cortisol level, salivary immunoglobulin A level, and a profile of mood states score. The sensation of appetite was assessed using visual analog scale scores, and fasting plasma acylated ghrelin, insulin, and glucose were measured. The cortisol concentrations were significantly higher at Camp2 (466.7 +/- 60.7 nmol . L-1) than at Rest (356.3 +/- 100.9 nmol .L-1; p = 0.002) or Regular (361.7 +/- 111.4 nmol . L-1; p = 0.003). Both prospective and actual food consumption significantly decreased at Camp2, and acylated ghrelin concentration was significantly lower at Camp1 (34.2 +/- 8.0 pg . m L-1) and Camp2 (32.0 +/- 8.7 pg . m L-1) than at Rest (47.2 +/- 11.2 pg . m L-1) or Regular (53.4 +/- 12.6 pg . m L-1). Furthermore, the change in acylated ghrelin level was negatively correlated with the change in cortisol concentration. This study's findings suggest that an early-phase physiological stress response may decrease the acylated ghrelin level in male power-trained athletes during an intensive training camp.

  • Ageing and free-living daily physical activity effects on salivary beta-defensin 2 secretion

    This study examined ageing and free-living daily physical activity effects on salivary human beta-defensin 2 (hBD2). A total of 168 healthy elderly and 26 healthy young volunteers underwent saliva sampling. Free-living step count, energy expenditure and activity durations at specific intensity levels (inactive, light, moderate and vigorous) were evaluated. The results show significantly lower salivary hBD2 secretion rates for elderly than for young participants (P&lt;0.05). Data from elderly participants were stratified by steps per day using quartiles (Q1-Q4) for distribution. Elderly in quartiles respectively showed step counts of 3145 +/- 129 in Q1, 5294 +/- 83 in Q2, 7001 +/- 86 in Q3 and 10,236 +/- 416 steps day(-1) in Q4. In elderly participants, significant differences were found in the mean step count, energy expenditure and activity duration with increasing pedometer-determined activity quartiles. hBD2 secretion rates were significantly higher for Q2, Q3 and Q4 than for Q1 (P&lt;0.05). Elderly participants in Q3 had the highest hBD2 secretion. In conclusion, these results suggest that moderate physical activity in daily living improves age-related impairment of oral immune function mediated by hBD2. For oral immune function enhancement and for prevention of upper respiratory tract infections in elderly people, we recommend accumulation of more than 7000 steps day(-1).

    This study examined changes in toll-like receptor 4 (TLR-4)-expressing monocytes and lymphocyte subpopulations in response to continuous intensive exercise training in athletes, as well as the effect of coenzyme Q10 (CoQ10) supplementation on these changes. Eighteen male elite kendo athletes in Japan were randomly assigned to a CoQ10-supplementation group (n = 9) or a placebo-supplementation group (n = 9) using a double-blind method. Subjects in the CoQ10 group took 300 mg CoQ10 per day for 20 days. Subjects in the placebo group took the same dosage of placebo. All subjects practiced kendo 5.5 h per day for 6 consecutive days during the study period. Blood samples were collected 2 weeks before training, on the first day (day 1), third day (day 3), and fifth day of training (day 5), and 1 week after the training period (post-training) to ascertain TLR-4(+)/CD14(+) monocyte and lymphocyte subpopulations (CD3(+), CD4(+), CD8(+), CD28(+)/CD4(+), CD28(+)/CD8(+), and CD56(+)/CD3(-) cells) using flow cytometry analysis. The group x time interaction for TLR-4(+)/CD14(+) cells did not reach significance (p = 0.08). Within the CoQ10 group, the absolute number of TLR-4(+)/CD14(+) cells was significantly higher only at day 5. The placebo group showed a significant increase in the absolute number of TLR-4(+)/CD14(+) cells at day 3, day 5, and post-training (p &lt; 0.05). There was no significant group x time interaction for any lymphocyte subpopulation. CD3(+), CD8(+), and CD56(+)/CD3(-) cells were significantly reduced at day 3 in both groups (p &lt; 0.05). In conclusion, CoQ10 supplementation might downregulate the increase of TLR-4-expressing monocytes in response to continuous strenuous exercise training in kendo athletes.

  • Use of Supplements by Japanese Elite Athletes for the 2012 Olympic Games in London

    Objective: To investigate supplement use among Japanese elite athletes.
    Design: We conducted a cross-sectional survey study using individual interviews during athletes' medical evaluations.
    Setting: One to 5 months before the Olympic Games in 2012, London, United Kingdom.
    Participants: Five hundred fifty-two Japanese athletes for the London Games, including candidates.
    Independent Variables: Sex, sports, supplement category, and participation.
    Main Outcome Measures: Whether athletes used supplements, what products were used, the frequency and purposes of use, and from what sources athletes received information on supplements.
    Results: All 552 athletes were interviewed by pharmacists regarding supplement use and agreed to the survey. Of them, 452 (81.9%) used 1 or more supplement products in the year before the study; a total of 952 different products (mean = 1.7, SD = 1.4) were used. The most commonly used supplement was amino acids (310 athletes; 56.2%), and 358 (38.7%) of the total products were amino acids. Of the athletes, 241 (43.7%) took at least 1 supplement daily, and of the supplements, 457 (49.4%) were taken daily. The most common purpose for supplement use was recovery from fatigue-327 (59.2%) athletes chose this answer, and 486 (52.5%) products were used for this purpose. Finally, regarding athletes' information sources on supplements, coaches, managers, and trainers were the most frequent advisors (275 athletes, 49.8%; 466 products, 50.4%).
    Conclusions: The results revealed widespread supplement use among Japanese elite athletes for the London Olympic Games. Education system was required not only for athletes but also for athletes' entourage, such as coaches, managers, trainers, and supplement companies.

Each subject continued intake of b240 or placebo and moderate-intensity resistance exercise for 12 weeks. Before and 4, 8, and 12 weeks after the start of intervention, each subject underwent saliva sampling. Before and after intervention, physical fitness tests and step count were measured. Our results showed that secretory immunoglobulin A secretion in 57 subjects during the b240/placebo intake period was significantly greater in the b240 group than in the placebo group (p&lt;0.05). There were no significant changes in physical fitness tests before and after intervention in the 2 groups. The daily amount of walking increased significantly after intervention in both groups (p&lt;0.05). These results suggest that in elderly adults with low physical activity and fitness, intake of b240 with appropriate physical exercise elevate salivary secretory immunoglobulin A secretion.

    Yoga stretching can be done comfortably and easily by beginners and older adults to compensate for lack of exercise or poor health maintenance. The aim of this study was to determine the effect of yoga stretching on mucosal immune functions, primarily human beta-defensin 2 (HBD-2) in saliva.
    Fifteen healthy adults (age, 60.4 +/- A 8.0 years) participated in the study. Participants rested for 90 min on the first day and performed yoga for 90 min on the second day. Measurements were carried out before and after rest or yoga. Saliva samples were collected by chewing a sterile cotton at a frequency of 60 cycles per min. Salivary HBD-2 concentration was measured using an enzyme-linked immunosorbent assay.
    HBD-2 concentration after yoga stretching (165.4 +/- A 127.1 pg/mL) was significantly higher than that before yoga stretching (84.1 +/- A 63.4 pg/mL; p &lt; 0.01). HBD-2 expression rate after yoga stretching (232.8 +/- A 192.9 pg/min) was significantly higher than that before yoga stretching (110.7 +/- A 96.8 pg/min; p &lt; 0.01). HBD-2 concentration (p &lt; 0.05) and HBD-2 expression rate (p &lt; 0.01) at post on the second day (yoga) was significantly higher than that on the first day (rest). POMS score of anger-hostility was lower after yoga than before.
    Yoga stretching for 90 min can increase salivary HBD-2 expression in older adults. Therefore, yoga stretching might be useful for older adults and athletes to maintain their health.

Secretory immunoglobulin A concentration on the forearm was significantly lower at B2 (p &lt; 0.05) and B3 (p &lt; 0.05) than that at B1 and that on the chest at B1 tended to be higher compared with B2 (p = 0.084) and B3 (p = 0.075). The number of staphylococci was significantly higher at B2 than that at B1 (p &lt; 0.01) and B4 (p &lt; 0.01) on the forearm. We conclude that high-intensity endurance exercise might depress immune function and enhance infectious risk on skin surface. High-intensity endurance exercise might depress the immune barrier and physical barrier and enhance the risk of skin infection. On the other hand, the biochemical barrier increases after exercise, and our findings suggest that this barrier might supplement the compromised function of other skin barriers.

    運動誘発性気管支喘息、気道過敏性を含む気管支喘息の有病率は一般人のみならず、アスリートにおいても増加している。先行研究で報告されている諸外国のトップアスリートの気管支喘息の有病率は、その検出する方法が統一されているものではないが、自施設における日本のトップアスリートの喘息有病率と比較して高い。アスリートのパフォーマンスやドーピング違反の視点から、トップアスリートに関わるスポーツドクターにとって正確な日本のトップアスリートの喘息有病率をしることは、非常に重要なことである。この論文の目的は喘息診断のためにどのような呼吸機能検査を適応するのか、そして日本のトップアスリートの喘息有病率がどの程度であるかを示すことである。喘息に関する質問表とスパイロメトリ検査をスクリーニング検査として北京オリンピック出場選手(候補選手を含む)482名全員に行った。気道可逆性テストおよびメサコリンや運動による誘発性テストも適宜行った。気道可逆性テストおよび誘発性テストで陽性になったのは6.2%(482名中30名)、無症状にも関わらず喘息と診断されたのは3.5%(482名中17名)であった。最終的に検査の結果やアスリートの希望により喘息の治療が施されたアスリートは12.4%(482名中60名)であった。この論文はスパイロメトリ検査をスクリーニング検査として施行し、客観的な検査を用いて日本オリンピックアスリートの喘息を診断した最初の報告である。この方法によって最終的に得られた日本のトップアスリートの喘息有病率は報告されている諸外国の有病率と相違ないものであった。(著者抄録)

Of these athletes, 47 (62.7%) used 1 or more supplement products (average number of products used, 1.1 +/- 1.3). The most popular supplement was amino acids, which were used by 33 athletes (44.0%). Of the supplements used, 28 (32.6%) were taken every day, whereas 28 (32.6%) were used only on special occasions. Moreover, 34 of the supplement products (39.5%) were taken to enhance recovery from fatigue, whereas 19 (22.1%) were used to improve athletic performance. Statistical analysis suggested that there was no significant relationship between supplement use and attitudes toward a balanced diet.
    Conclusions: The results revealed widespread supplement use among young elite athletes in the Japanese national team for the Singapore Youth Olympic Games in 2010. Moreover, these athletes apparently used supplements without considering the effects of their normal diets.

Subjects reported the appearance of URTI symptoms (sore throat, headache, runny nose, coughing, or fever), if any, during the prior month. The serum estradiol concentration and salivary SIgA secretion rate were significantly lower for ARs than for ERs (p &lt; 0.05). Serum progesterone concentration was not significantly different between groups. Higher frequencies of headache, runny nose, coughing, and fever were observed in ARs than in ERs. Results show that athletic amenorrhea with low estrogen might accelerate downregulation of mucosal immune function in athletes and enhance susceptibility to infection.

Subjects in CON maintained their normal physical activity levels during the study period. Blood samples were collected before and after the training period. Samples were measured for the numbers of leukocytes, lymphocytes, and monocytes, and for CD3(+), CD4(+), CD8(+), CD28(+) CD4(+), CD28(+) CD8(+), TRL-4(+) CD14(+), and CD80(+)CD14(+) cells. The number of leukocytes, lymphocytes, monocytes, CD3(+), CD4(+), and CD8(+) cells did not change after 12 weeks in either EXC or CON. The number of CD28(+)CD8(+) cells increased significantly after training in EXC (p &lt;= 0.05), although CON showed no significant change. In the EXC group, CD80(+)CD14(+) cell counts were significantly higher after training (p &lt;= 0.05), but the TLR-4(+)CD14(+) cell counts were unchanged. In the CON group, no significant alteration existed in TLR-4(+)CD14(+) and CD80(+)CD14(+) cell numbers. In conclusion, exercise training in elderly people is associated with increased CD28-expressing Tc cells and CD80-expressing monocytes. Therefore, exercise training might upregulate monocyte and T-cell-mediated immunity in elderly people.

The CD3(+), CD4(+), CD8(+), and CD28(+)CD4(+) cell counts were significantly lower at WL than at BL (p &lt;= 0.05); they reverted to the baseline value at AC. The TLR-4(+)CD14(+) cells were significantly fewer at WL (p &lt;= 0.05); they remained fewer than they had been at BL, even at AC. These results suggest that 2 weeks of weight loss before a competition can impair cell-mediated immune function and induce high susceptibility to URTI in judo athletes. Coaches, support staff, and athletes should monitor athletes&apos; weight loss, hydration status, appearance of URTI symptoms, and immunocompetence such as lymphocytes and monocytes to prevent the physical condition from becoming worse.

    Epidemiological studies suggest that highly trained athletes are more susceptible to upper respiratory tract infections (URTI) compared with the general population. Upper respiratory symptoms (URS) often appear as either primary invasion of pathogenic organisms and/or reactivation of latent viruses such as Epstein-Barr virus (EBV). The purpose of this study was to examine the relationship between EBV reactivation and the appearance of URS during intensive training in collegiate rugby football players. We evaluated EBV-DNA expression in saliva and examined the relationship between onset of URS and daily changes in EBV-DNA as well as secretory immunoglobulin A (SIgA) levels among 32 male collegiate rugby football players during a 1-month training camp. The EBV-DNA expression tended to be higher in subjects who exhibited sore throat (p = 0.07) and cough (p = 0.18) than that of those who had no symptoms, although their differences were not significant. The SIgA level was significantly lower 1 day before the EBV-DNA expression (p &lt; 0.05). The number of URS increased along with the EBV-DNA expression and decrease of SIgA levels. These results suggest that the appearance of URS is associated with reactivation of EBV and reduction of SIgA during training.

    We aimed to develop a valid model of immunosuppression induced by intense exercise in rats. Rats were divided into three groups. In the rest (Rest) group, saliva was collected from resting rats on 4 consecutive days. In the exercise (Ex) group, rats ran on a treadmill untill exhaustion (exercise time: 60.0 +/- 3.7 min), and their saliva was collected before and after exercise; the salivary glands were removed after exercise. In the control (Con) group, saliva collection and gland removal were also performed, but the rats did not exercise. Secretory immunoglobulin A (SIgA) concentrations in saliva and polymeric immunoglobulin receptor (pIgR) mRNA expression in the glands were measured. There was no significant change in SIgA concentration in the Rest group over 4 days. In the Ex group, SIgA concentration decreased significantly after exercise compared with before, whereas there was no significant change in the Con group. The expression of pIgR mRNA was significantly lower in the Ex group post-exercise than in the Con group. Our procedure for saliva collection appeared suitable, and the exercise-induced SIgA suppression was probably caused by a decline in pIgR mRNA expression. We propose to use this reproducible and reliable rat model of exercise-induced SIgA suppression in future studies.

    CD28 molecule expression on the surface of T cells plays a critical role in up-regulation of various cytokines synthesis and T-helper (Th) cell proliferation and differentiation. However aging induces a decrease in CD28 expression and unbalance of Th1/Th2, leading to impairment of Th-cell mediated immune function. The purpose of this study was to assess the affects of moderate exercise training on CD28 expression and the balance of Th1/Th2 cells in elderly people. Forty-eight elderly subjects were assigned to an exercise training group (EXC: 13 males, 15 females; aged 61-76) or a non-exercise control group (CON: 7 males, 13 females; aged 62-79). Subjects in EXC participated in exercise sessions 5-days a week for 6 months. Meanwhile, subjects in CON maintained their normal physical activity levels during the study period. Blood samples were collected before and after the training period. Samples were measured for the number of leukocytes and lymphocytes, as well as for CD3(+), CD4(+), CD28(+)CD4(+), IFN-gamma(+)CD4(+), IL-4(+)CD4(+) cells. The number of leukocytes, lymphocytes, and CD3+ cells did not change after 6 months in both EXC and CON. The number of CD4(+) and CD28(+)CD4(+) cells significantly increased after the training in EXC (P &lt; 0.05), while CON did not show significant changes. In the EXC group, IFN-gamma(+)CD4(+) cell numbers were significantly higher following the training (P &lt; 0.05), but the number of IL-4(+)CD4(+) cells was not changed. In the CON group, there were no significant alterations in IFN-gamma(+)CD4(+) and IL-4(+)CD4(+) cell numbers. In conclusion, moderate exercise training in the elderly is associated with improvement of expression of CD28 on Th cells and Th1/Th2 balances. Therefore, exercise training could up-regulate Th cell-mediated immune functions and be helpful for a decrease in the risk of infections and autoimmune diseases in elderly people.

    Purpose: The aim of this work was to assess the relationship in elderly subjects between free-living daily physical activity and mucosal immunity, especially salivary secretory immunoglobulin A (SIgA). Methods: Elderly volunteers (114 men and 170 women) aged 71.3 +/- 0.3 yr (range: 65-86 yr) participated in this study. Resting saliva samples were collected in the morning. Saliva samples stimulated by chewing a sterile cotton ball at a frequency of 60/60 s were collected. The SIgA concentration was measured using enzyme-linked immunosorbent assay (ELISA), and the SIgA secretion rate was calculated. Free-living step count (steps per day), energy expenditure (kJ(.)kg(-1)-d(-1)), and activity durations (min(.)d(-1)) at specific intensity levels (inactive, light, moderate, and vigorous) were evaluated using an electric pedometer. The data obtained were stratified by pedometer-determined steps per day using quartiles (Q1-Q4) for distribution. Results: Elderly in quartiles showed step counts of 2962 94, 5118 62, 6832 59, and 9951 264 steps per day. Significant differences were found in the mean step count (P &lt; 0.0001), energy expenditure (P &lt; 0.0001), and activity duration (P &lt; 0.0001) with increasing pedometer-determined activity quartiles. Both SIgA concentration and SIgA-secretion rate were significantly higher for Q3 than for Q1 (P &lt; 0.05). Meanwhile, saliva flow rates showed no significant differences across quartiles. Conclusion: These results suggest that a free-living daily physical activity level of approximately 7000 steps per day might be regarded as a moderate daily physical activity target for elderly people to improve mucosal immune function.

    The influence of age and gender on salivary secretory immunoglobulin A (SIgA) in response to moderate exercise training was studied in 158 elderly subjects, Subjects were assigned to an exercise training group (EXC: 51 males, 74 females) or a non-exercise control group (CON: 11 males, 22 females). The subjects in each group were separated into four age-gender subgroups (60-69-yr-old males, over 70-yr-old males, 60-69-yr-old females, over 70-yr-old females) and compared by age and gender Subjects in EXC participated in exercise sessions 5-days a week for 6 months. Saliva samples were collected both before and after the study period. The SIgA secretion rates were significantly increased after training (p < 0.05) in all the age-gender subgroups of EXC (60-69 males: 41%, over 70 males: 55%, 60-69 females: 40%, over 70 females: 38%); no age- or gender-related differences were observed. On the other hand, none of the age-gender subgroups of CON showed significant changes in the SIgA secretion rate; also, there were no age- or gender-related differences. In conclusion, enhancement of mucosal immune function following regular moderate exercise training occurs in elderlies in their 60s and over 70 years, and in both, males and females.

    Background: The immune system declines in efficiency with advancing age, making the elderly less resistant to pathogenic microorganisms. Upper respiratory tract infection (URTI) is a common illness. Recent studies have shown that suppression of secretory immunoglobulin A (SIgA) is associated with increased incidence of URTL
    Objective: To assess the effect of exercise on salivary SIgA in elderly subjects.
    Methods: Forty five elderly subjects (18 men, 27 women; mean (SD) age 64.9 (8.4) years) performed both 60 minute resistance and 60 minute moderate endurance training a week for 12 months. Saliva samples were obtained before training, and at four and 12 months during the training period. Salivary SIgA concentrations were measured by enzyme linked immunosorbent assay, and the SIgA secretion rate was calculated.
    Results: SlgA concentrations before training, and at four and 12 months during training were 24.7 (14.4), 27.2 (14.2), and 33.8 (18.5) mug/ml respectively. SIgA secretion rates were 29.5 (26.0), 33.8 (27.2) and 46.5 (35.1) mug/min respectively. The results indicate that both the concentration and secretion rate of SlgA significantly (p&lt;0.01) increased during 12 months of exercise in these elderly subjects.
    Conclusion: Regular moderate exercise seems to enhance mucosal immune function in elderly subjects.

    Intercellular adhesion molecule-1 (ICAM-1) plays an important role in leukocyte migration from the circulation and intervention at sites of inflammation. We investigated the effects of various types of exercise on circulating levels of soluble ICAM-1 (sICAM-1) in normal healthy male adults. Plasma concentrations of sICAM-1 were measured before and after bicycle ergometer exercise at intensity of 80% maximal oxygen consumption (VO2max) (16 min), 42 km endurance running and 30-min downhill running at intensity of ventilation threshold (VT). The plasma sICAM-1 level increased 1 day after the endurance running (12%) and downhill running (14%), but not after ergometer exercise. Plasma C-reactive protein (CRP) and creatine kinase (CK) concentrations also increased 1 day after running. Our data suggest that exercise associated with muscle damage and/or inflammation results in increased levels of plasma sICAM-1. The physiological significance of post-exercise high plasma sICAM-1 levels is not clear at this stage, but changes in plasma sICAM-1 may reflect the status of the immune system.

    PURPOSE: We examined the levels of autoantibodies against prethrombin-1 and fragment-1 in pregnant women to determine the type of autoantibodies that were associated with severe, preeclampsia or spontaneous abortion.
    SUBJECTS AND METHODS: We measured autoantibodies bound to prethrombin-1, prethromhin-1, and fragment-1 by using an enzyme-linked immunosorbent assay (ELISA) in 12 healthy nonpregnant women, 36 women with normal pregnancies, 28 pregnant women with severe preeclampsia, and 19 pregnant women who subsequently had spontaneous abortion.
    RESULTS: Plasma samples in 10 (36%) of the 28 women with severe preeclampsia and 11 (58%) of the 19 women with spontaneous abortion were positive for antiprothrombin antibodies as compared with 3 (9%) of the 36 women with normal pregnancies. AU II of the positive samples from women who had spontaneous abortions were positive for antiprethrombin-1 antibody, but only 1 was positive for antifragment-1 antibody. The mean ( +/- SD) titer of antiprethrombin-1 1 antibodies in patients with spontaneous abortion (36 +/- 9 U) was higher than that in women with normal pregnancies (10 +/- 4 U; P &lt; 0.01). Antiprethrombin-1 antibody was detected in only 28 women with severe preeclampsia, whereas all 10 women with antiprethrombin antibodies were positive for antifragment-1 antibody. The antifragment-1 antibody titer in patients with severe pre eclampsia (49 +/- 15 U) was higher than in women with normal pregnancies(l3 +/- 6 U, P &lt;0.01).
    CONCLUSIONS: There is a strong and specific association between various types of antiprothrombin antibodies with severe preeclampsia and spontaneous abortion. (C)2001 by Excerpta Medica, Inc.

    Palmar fasciitis and polyarthritis (PFA) is a rare paraneoplastic rheumatic syndrome characterized by flexion contractures of both hands and thickening of palmar fascia. Several reports have suggested that this syndrome is a tumor-associated autoimmune disorder. We report a 44-year-old Japanese man who presented with flexion contractures of both hands associated with thickening of palmar fascia and polyarthritis. These clinical pictures were suggestive of PFA associated with occult neoplasm. Upper gastrointestinal endoscopic examination revealed advanced gastric cancer. Resection of the cancer resulted in a gradual resolution of palmar fasciitis and polyarthritis. This clinical course suggests an underlying tumor-related immunologic process in this syndrome.

  • The relationship between serum immunoglobulin levels and pulmonary involvement in systemic sclerosis

    Objective. To examine the relationship between serum immunoglobulin (Ig) levels and pulmonary function in patients with systemic sclerosis (SSc).
    Methods. Twenty-four patients with SSc who had at least 2 sets of pulmonary function tests (PFT) at intervals of more than one year were eligible. Multiple linear regression models were constructed for prediction of the annualized rates of change of forced vital capacity (FVC), carbon monoxide diffusing capacity (DLco), and DLco per unit alveolar volume (K-co).
    Results. The rates of change of FVC and K-co correlated with the annualized rate of change of IgG (p &lt; 0.001 and p = 0.005, respectively), and the rate of change of DLco correlated with the serum IgM level at the first PFT (p = 0.020) and with the annualized rate of change of IgG (p = 0.007).
    Conclusion. The rates of change of serum Ig levels are associated with those of pulmonary function in SSc. Use of this model may assist investigation of pulmonary involvement.

  • Effect of brief maximal exercise on circulating levels of interleukin-12

    Interleukin-12 (IL-12) is a cytokine that was originally identified as natural killer cell stimulatory factor. It induces the activity of T-helper 1 (Th1) cells and exhibits strong anti-tumor activity. In this study, we studied the effects of brief anaerobic maximal exercise on circulating levels of IL-12. Six healthy males [mean (SD) 25.2 (2.6) years] performed a modified Wingate test exercise (resistance 0.075 kg/kg of body mass). The exercise consisted of five bouts of maximal cycling for 10 s, with rest intervals of 50 s between them. Blood samples were taken before, immediately after, 30 min after, 60 min after and 120 min after the exercise. Plasma concentrations of IL-12 were measured using an enzyme-linked immunosorbent assay. Data were corrected for hemoglobin and hematocrit measurements. Plasma concentrations of IL-12 averaged [mean (SD)] 234.2 (40.9) pg/ml before, 305.2 (62.1) pg/ml immediately after, 202.8 (24.2) pg/ml 30 min after, 239.7 (35.1) pg/ml 60 min after, and 199.6 (49.2) pg/ml 120 min after the exercise. We showed that plasma concentrations of IL-12 increased significantly immediately after brief anaerobic maximal cycle ergometer exercise (P &lt; 0.01).

  • CHANGES IN NOCTURNAL URINARY CATECHOIAMINES EXCRETION AND AEROBIC FITNESS DURING KANGEIKO IN FEMALE COLLEGE KENDO PLAYERS

    Most kendo players have gone through a traditional form of training, called Kangeiko. Kangeiko is a high intersity training camp carried out during the cold early mornings of winter. The aim of Kangeiko is to cultivate the mind and improve the physical fitness of the kendo players. In this study, we examined the changes in nocturnal urinary catecholamines excretion, aerobic fitness, and subjective condition during kangeiko in kendo. Training in Kangeiko was performed for about two hours from 5: 00 to 7: 00 am., and Kangeiko was continued for ten days in January in 1995. Training program during Kangeiko consisted of Kirikaeshi and Kakarigeiko for 30-40minutes, and Gokakugeiko for about 30minutes by interval training form. We measured nocturnal urinary catecholamines (dopamine, adrenaline, and noradrenaline) excretion in five female college kendo players throughout Kangeiko, and evaluated their aerobic fitness before and the 3rd day after Kangeiko using multistage fitness test. Resting blood creatine kinase (CK) activity was measured before training on the first,6th, and 10th days of Kangeiko, and the 3rd day after Kangeiko. Each players kept recording their subjective condition, such as fatigue, muscle soreness etc., using 7-point rating scale from 1 (very bad) to 7 (very well) throughout Kangeiko. Nocturnal urinary catecholamines excretion were increased on the 3rd and/or 8th days of Kangeiko, but with no significance. Aerobic fitness fell the 3rd day after Kangeiko. There were significant correlations between changes in nocturnal urinary catecholamines excretion and aerobic fitness. Subjective condition was significantly decreased throughout Kangeiko. In most of the players, some of subjective condition became worse with the changes in nocturnal urinary catecholamines excretion. Compared with the first day of Kangeiko, resting blood CK activities were significantly increased on the 6th and 10th days of Kangeiko, indicated that repetition of high intensity training during Kangeiko produced remarkable disruption of muscle tissue. Change in basal sympathetic activity induced by extreme physical and mindful stress during Kangeiko might cause changes in aerobic fitness and subjective condition of the kendo players. Training program should be designed in consideration of individual differences in those stress during Kangeiko, as well as adequate health care.

    The purpose of this study was to investigate the effects of &lt;i&gt;Lactobacillus gasseri&lt;/i&gt; OLL2809 (MG2809) intake on salivary secretory immunoglobulin A (SIgA) secretion and incidence of upper-respiratory tract infection (URTI) symptoms among college rugby athletes. Sixty-seven subjects were assigned to a MG2809 group (n = 33) or a placebo group (n = 34) using a double-blind procedure. Each subject took MG2809 or placebo tablets for 9 weeks. Resting saliva samples were collected before (0 week) and after 2, 4, 6, 8, and 9 weeks of the MG2809 intakes. A log of URTI symptoms was kept every day during the study period. Our results showed that salivary SIgA level was significantly increased at 6 weeks in the MG2809 group (p &lt; 0.05), although the placebo group did not show significant change. The duration of URTI symptoms episode in the MG2809 group was significantly shorter than that in the placebo (p &lt; 0.05). These results suggest that regular intake of MG2809 may enhance oral immune function mediated by SIgA and reduce the risk of URTI in athletes.

    &lt;b&gt;Objective:&lt;/b&gt; The purpose of this study was to examine the effects of Coenzyme Q10 (CoQ10) supplementation on salivary secretory immunoglobulin A (SIgA) and health-related Quality of Life (QOL) in middle-aged and elderly individuals. &lt;b&gt;Methods:&lt;/b&gt; Sixty healthy middle-aged and elderly individuals were assigned randomly to a CoQ10 supplementation group (n = 30) or a placebo supplementation group (n = 30) using a double blind method. Subjects took 150 mg CoQ10 or placebo per day for 8 weeks. Salivary SIgA secretion rate and SF-36 test (physical and mental health-related QOL) were determined before and after the intervention. &lt;b&gt;Results:&lt;/b&gt; CoQ10 group showed that a tendency to increase of salivary SIgA secretion rate (p = 0.08), although placebo group did not show significant change. Physical health-related QOL did not significantly change in both groups. Vitality and mental health scores, which were subscale of mental health-related QOL, were significantly increased after the intervention in CoQ10 group (p &lt; 0.05), although placebo group did not show significant change. &lt;b&gt;Conclusion:&lt;/b&gt; 8 weeks of CoQ10 supplementation may bolster SIgA-mediated oral immunity and mental health-related QOL in middle-aged and elderly individuals.

    In this study, to investigate the influence on the innate immune function due to the execution time zone of continuous exercise training, we studied the relationship with the amount of physical activity (Total consumption, momentum, step count, stride, activity time) and body composition (Height, body weight, body fat mass, muscle mass, protein mass) and innate immune function (number of dendritic cells and dendric cells + toll like receptor 4) along with the execution time zone of training. As a result, although it was not possible to clarify the effect of the innate immune function depending on the execution time zone of training, it was suggested that innate immunity declines due to aging. On the other hand, it was suggested that raising muscle mass might maintain or improve innate immune function regardless of aging. In the future, it is necessary to investigate the relationship between training intervention incorporating low load resistance training and innate immune function

    The purpose of this study was evaluating the availability of salivary HSP70 to assess the physical condition in athletes and elderly individuals. The salivary HSP70 was increased after the acute intensive exercise. In athletes, continuous intensive exercise did not markedly change resting salivary HSP70 levels. This result might be influenced by individual differences of HSP70 responses to intensive exercise. In elderly subjects, continuous moderate exercise training enhanced resting salivary HSP70. In addition, HSP70 might be involved with T-cell activation in response to exercise training. In conclusion, salivary HSP70 might be useful to assess the physical condition in elderly subjects, while it might require to consideration in athletes

  • 新しい自己抗体,抗ILー1α抗体の定量的測定法の確立と臨床的意義に関する研究

     View Summary

    抗ILー1α自己抗体の定量的測定法を開発して、リウマチ性疾患の多数の症例で測定した。我々が開発した方法は、proteinAセファロ-スに一定量の血清IgGを結合させ、 ^<125>IーILー1αと反応させることによって抗体と結合したILー1αの量をガンマカウンタ-によって測定した。その結果、正常人では抗ILー1α自己抗体陽性血清の頻度は約5%であるのに対し、慢性関節リウマチ疾者では約15%と3倍の陽性頻度を示した。全身性エリテマト-デス患者や強皮症患者では、ほぼ正常人と同程度の陽性頻度であった。リウマチ性滑膜炎と抗ILー1α自己抗体の出現の関係を明らかにするため、数人の患者について、2ー3年にわたって血清の抗ILー1α抗体の抗体価と疾患活動性との関係を調べた。その結果、大部分の症例では抗ILー1α抗体価は滑膜炎の程度と相関した。この結果は、リウマチ性滑膜炎において出現するILー1αが、抗原として作用し、抗体産生を誘導したことを示唆する。しかし、正常人でも5%とかなり高い頻度でこの自己抗体が出現することから、最初の抗原刺激がILーαそのものであるかどうかは、はっきりしない。そこで、この自己抗体が認識するILー1α分子上のエピト-プを、βガラクトシダ-ゼとILー1αの融合蛋白を大腸菌に作成させ、抗体との反応性を検討することにより決定することを試みた。しかし、融合蛋白の精製の過程でILー1α部分を含む蛋白の変性がおこってしまうこと、それにより、変性ILー1αが非特異的にFc部分を介してIgGに結合してしまうことがわかり、この方法ではエピト-プの決定ができないことが判明した。そこで、現在、ILー1αの色々な部分の合成ペプチドを作成し、これらと抗ILー1αとの反応性を調べることによりエピト-プの決定を試みている

  • 運動が免疫機能にあたえる影響に関する研究

     View Summary

    運動が免疫機能に与える影響を知るために、細胞生免疫機能とくにウイルス感染症や癌の防御に重要な役割を果たしているNatural Killer細胞(NK細胞)に焦点を絞り検討した。自転車エルゴメ-タ-を用いた急性運動負荷により、ヒトNK細胞活性は負荷直後より急激に上昇し、運動終了後は運動前の値より有意の低値を示した。この運動終了後のNK細胞活性の低下は徐々に前値に戻りおよそ6時間後には回復した。運動習慣によるNK細胞活性への影響を検討するために陸上長距離選手のNK細胞活性を検討したところ、長距離選手ではNK細胞活性が高値であることが明らかとなった。そこでこの高値が恒常的であるか否かを知るために、13名の陸上長距離選手のNK細胞活性の変動をトレ-ニング開始前から縦断的に測定し検討した。トレ-ニング開始前のNK細胞活性は持久的トレ-ニングを持続するに従い徐々に増加した。また、トレ-ニング期間中に実施した2回の強化合宿によりNK細胞活性はさらに一段と上昇した。強化合宿終了直後は若干低下傾向がみられたものの、全体としとはトレ-ニング期間中を通じてNK細胞活性は高値に保たれた。以上の事から急性運動負荷そして運動習慣とくに持久的運動がNK細胞活性を上昇させることが明らかとなった。この運動によるNK細胞活性上昇の機序また上昇したNK細胞活性が臨床的な感染防御にどのような働きをするかなどについてさらに詳細の研究を要すると考えられた

  • 有酸素運動が免疫機能にあたえる影響に関する研究

    運動が健康の増進・維持に効果があるという考えが広く受け入れられるようになり、健康スポーツがますます盛んとなっている。本研究では健康の維持・増進に有効とされている有酸素運動が免疫機能とくに細胞性免疫機能に与える影響を検討した。有酸素運動の習慣をもつ成人では細胞性免疫機能とくにナチュラルキラー細胞(NK細胞)の活性が運動習慣のないものと比較すると男女とも高いことが明らかとなった。また、経時的にNK細胞活性の変動をみると、日々の運動量が増すに従い活性が高値を示していくことも明らかとなった。つぎに、このような有酸素運動がNK細胞活性へ与える影響の機序を探る目的でNK細胞にレセプターを持つカテコラミンなどのホルモンとの関係を検討した。急性の有酸素運動負荷により、各種ホルモンがNK細胞活性の変動の平行して変動することが明らかとなった。また、マウスを用いた実験系ではカテコラミンの共存下ではその濃度によりNK細胞の活性が影響を受けることも示唆された。かかる成績から有酸素運動が免疫に与える影響の機序には複数の因子が関与していると考えられた。また、情報伝達物質であるサイトカインの関与についても検討したが、運動により変動するという結果を得たのみで機序への関与については今後の課題となった。今後運動と免疫に関する研究をさらに進めるためには情報伝達物質の関与を含め様々な視点からのアプローチが必要と考えられた

  • 免疫系に及ぼす運動の影響に関する研究:情報伝達物質と介する機構について

    免疫系におよぼす運動の影響に関与すると考えられる情報伝達物質としてサイトカインに着目し研究を進めた。単球/マクロファージに関連するインターロイキン1β、腫瘍壊死因子、インターロイキン6、可溶性インターロイキン2レセプターについて健常者5名を対象とし、65%VO_2maxで30分間の自転車エルゴメーター負荷を行い、その変動を検討した。サイトカインの測定は、血漿サイトカイン濃度と、血球のサイトカイン産生能(全血培養法による)について実施した。血漿サイトカイン濃度ではIL-6濃度は負荷直後に増加傾向、SIL-2r濃度は負荷後に減少傾向を示したが、IL-1βとTNFα濃度は低値のため測定不可能であった。血球サイトカイン産生では負荷直後、IL-1βの産生が増加したものに、TNFαの産生も運動によって亢進することが示唆された。生体の状況に近い全血培養法による測定でIL-1βとTNFαの産生に増加が認められた。このことから、運動が免疫機能に与える影響の背景にはこのようなサイトカインが関与していることが示唆された。今後さらにサイトカインの作用する部位についての研究を進めたい

  • ループス抗凝固因子の対応抗原に関する研究

     View Summary

    1.ループス抗凝固因子と抗beta_2-glycoprotein I抗体との関係全身性エリテマトーデス患者96例の血清において、活性部分トロンボプラスチン時間が延長している15例のうち9例は抗beta_2-glycoprotein I抗体陽性であったが、6例は陰性であった。また、抗beta_2-glycoprotein I抗体陽性の33例中で活性部分トロンボプラスチン時間が正常であったのは22例であった。2.ルーブス抗凝固因子の対応抗原の検索まず、トロンボモジュリンについて検討した。固相化した抗トロンボモジュリン・モノクローナル抗体にトロンボモジュリンを結合させ、それを抗原として血清中の抗トロンボモジュリン抗体を酵素免疫測定法で検出しようと試みた。しかし、ループス抗凝固因子陽性血清および陰性血清で明らかな差異は認めなかった。次にプロトロンビンについて検討した。プロトロンビンを固相化して血漿中の抗プロトロンビン抗体を反応させ、それを酵素免疫測定法により検出した。ループス抗凝固因子陽性の血漿2例中2例で抗プロトロンビン抗体が検出された。ループス抗凝固因子陰性の血漿2例では抗プロトロンビン抗体は検出されなかった。また、固相化したプロトロンビンに対する反応はカルシウムイオン存在下でわずかに減少する傾向があった。陰性荷電のリン脂質としてカルジオリピンを固相化し、カルシウムイオン存在下でプロトロンビンを反応させて、さらに血漿中の抗プロトロンビン抗体を反応させてみたが、抗プロトロンビン抗体は検出できなかった。今後、検出された抗体が抗プロトロンビン抗体であることをさらに確認するとともに、ループス抗凝固因子がすべて抗プロトロンビン抗体として説明されるか否かを検討していく予定である

  • 運動による免疫機能の変化.サイトカインを介する制御機構に関する研究

    健康な成人を対象として運動負荷を行ないリンパ球機能、NK細胞機能、単球機能、などの免疫機能を測定すると同時に、インターロイキン、TNF、インターフェロンなどのサイトカインを分析した。この結果、運動により変化する免疫機能の内リンパ球やNK細胞活性などの変動に伴い、TNFなどのサイトカインおよびそのレセプターが変動することを示唆する成績を得た。この結果は免疫系の情報伝達物質として最も重要な役割を担っているとされるサイトカインの動態に運動が影響を与えことを反映する結果と推察され、運動による免疫系の変化の制御にサイトカインが関わっている可能性は高いと考えられた。現在、細胞培養系を用いてin vitroでの検討を進める段階にいたっている。また、近年運動との関係が注目されている活性酸素も免疫を含めた生体防御機構に関連が深い。このことから運動による免疫系の変化に及ぼす活性酸素の影響をサイトカインとの関連を考慮しつつ検討した。この結果好中球の産生する活性酸素種は急性の運動負荷では負荷直後に上昇しその後低下するなどNK細胞活性に近い変動パタ-を示すことが明らかとなった。また、長時間の有酸素運動負荷では運動終了後も数日にわたって上昇を続ける傾向を認めた。この活性酸素種の産制にも情報伝達物質が関与しているとの報告もあり、運動による免疫機能変化の機序にはサイトカインを含めた情報伝達物質の関与が強く示唆された

  • EFFECT OF ECERCISE ON HUMAN DEFENCE MECHANISMS

    We investigated the effect of physical exercise on human defense mechanisms. Firstly we surveyed the recent research tendency focusing on the change of immune parameters after physical exercise. We found production of superoxide was influenced by exercise. We studied the usefulness of secretary IgA in saliva as, an indicator of human defense system. As saliva is obtained non-invasively, it might be a good tool to evaluate the human defense system in sports field. After intensive training camp, the IgA was elicited to decrease and NK cell activities was also found to decrease temporarily. We examined the change of cytokines after physical exercise. Interleukin 2 receptor and interleukin 6 receptor was found to decrease in plasma. Cytokines might play important role in the mechanisms of change of immune system by exersise stress. These results of this research might suggest that appropriate exercise enhance the human defense system

  • Exercise induced alteration of secretory immunoglobulin A

    1. Levels of secretory immunoglobulin A (sIgA) and risk of upper respiratory tract infection (URTI). Subjects were 31 men. Levels of sIgA in saliva were measured and the incidence of URTI was investigated for two months. Six persons were taken URTI.Some of them were taken URTI after decrease of sIgA.Further investigation is needed, because few people were taken URTI.2. Amount of exercise and level of sIgA(1) Nine boys (age 10-12 years old) were checked with the 20-m shuttle run test. They ran for 7.7±1.5 minutes in the test. The secretion rate of sIgA in saliva was 20.2±3.1 μg/min before the test, and 15.5±3.2 μg/min after the test. There was no significant difference between before and after the test. It was suggested that short maximum exercise did not reduce the level of sIgA in saliva.(2) The sIgA secretion of the training group was compared with the sedentary control group. The sIgA secretion rate of the training group was 27.5±15.5 μg/min , and that of the sedentary control group was 19.9±14.9 μg/min. The training group had significantly more secretion of sIgA than the sedentary control group. Training might increase secretion of sIgA.3. Mechanism of post-exercise decrease of sIgAWe exercised mice, and collected saliva of mice before and after exercise. Four of five mice showed decrease of sIgA after exercise. Mice might be available to examine the mechanism of post-exercise decrease of sIgA

  • Development of a SIgA measurement kit for monitoring of risk of URTI

  • Effects of exercise on protection factors for upper respiratory tract infection.

  • The effects of exercise on immune senescence in elderly

    The purpose of this study was to examine the effects of exercise training on immune function in elderly individuals. Moderate exercise training might up-regulate T-cell activation in elderly. In older rat, moderate exercise training might increase splenic lymphocyte-subsets in relation to T-cell activation. Moreover, combined lactobacillus intake and moderate exercise training might bolster mucosal immune function in elderly subjects.   • 高強度運動による皮膚バリア機能低下の機序の解明

    本研究では、健常成人男性8名を対象に、高強度持久性運動が皮膚の物理的バリア関連因子に及ぼす影響を検討した。その結果、血中の脳由来神経栄養因子(Brain-derived neurotrophic factor; BDNF)とthymus and activation-regulated chemokine(TARC)が運動前と比較して運動直後に増加傾向を示し、運動直後と比較して運動30分後に有意な低下を示した。本結果から高強度運動直後のかゆみや炎症の増悪が示唆されたが、運動30分後には運動前と同等まで低下しているため、健常人の皮膚状態としては大きな変化は考えにくい。一方で、アトピー性皮膚炎患者では、高強度運動による皮膚炎症の増悪は重大な問題であり、今後は継続的な高強度運動による影響やアトピー性皮膚炎患者を対象とした研究が必要である。

    本年度は、実際のスポーツ現場でアスリートの皮膚バリア機能評価を実施する前段階として、皮膚バリア機能が皮膚表面の黄色ブドウ球菌の増殖に与える影響を検討することとした。皮膚モデルとしてヒト3次元培養表皮LabCyte EPI-MODEL (Japan Tissue Engineering)を使用し、分泌型免疫グロブリンA (secretory immunoglobulin A; SIgA)、ディフェンシン(human β-defensin 2; HBD-2)、コアグラーゼ陰性ブドウ球菌(coagulase-negative staphylococcus; CNS)および角質層の厚さによる黄色ブドウ球菌数や生細胞数の変動を検討する予定であった。しかしながら、ヒト3次元培養表皮を用いて細菌感染状態を検討した研究はなく、予備実験を実施し、プロトコールを確立する必要があった。実際にヒト3次元培養表皮を培養したのち、RNAやATPを測定し、生細胞数を計測した。また、培養された角質層の物理的バリア機能を評価するために皮膚透過性試験を行い、角質層のタイトジャンクションを確認した。これらの予備実験の結果から、皮膚バリア機能と黄色ブドウ球菌の増殖を検討するための具体的な研究プロトコールを確立した。また、身体活動量が皮膚バリア機能に及ぼす影響も検討した。健康な高齢者を対象とし、日常生活における身体活動量の違いが皮膚の物理的バリア機能に及ぼす影響を検討した。測定項目は、角質水分量、経皮水分蒸散量(trans-epidermal water loss; TEWL)、身体活動量とした。その結果、年齢と運動量(r=-0.406, p<0.01)、歩数(r=-0.465, p<0.01)が負の相関関係を示すことが確認され、目尻の角質水分量とTEWLに有意な負の相関関係がみられた(r=-0.326, p<0.01)。また、前期高齢者の目尻においては、歩数と角質水分量が負の相関関係を示し(r=-0.476, p<0.01)、歩数とTEWLが正の相関関係を示すことが明らかになった(r=0.365, p<0.05)。従って、前期高齢者において過度な身体活動が皮膚の物理的バリア機能を低下させる可能性が考えられるが、前腕における角質水分量およびTEWLには歩数との有意な相関関係が認められなかった。故に、ウォーキングをする際の紫外線の暴露が皮膚の物理的バリア機能を低下させると推察されるが、本研究では紫外線の暴露について検討していないため、今後さらに詳細に検討していく必要がある。

  • ドーピング禁止薬物の治療目的使用に関する研究

    (1)治療目的使用に係る除外措置申請の現状分析ドーピング禁止物質を疾患治療のため使用する競技者は「治療目的使用に係る除外措置(Therapeutic Use Exemption : TUE)」申請を行い、承認を得なければならない。国内のTUE申請数は、2010年度(2010年4月-2月末)で140件あった。(2)呼気凝縮液の炎症関連物質測定(目的)気管支喘息の客観的な診断は、気道可逆性試験や気道過敏性試験としてスパイロメトリーで気管支収縮を間接的に測定する方法が重視されているが、検査に長時間を要し、気管支喘息の一時的悪化の危険性も伴うため、競技者の運動誘発性気管支れん縮の診断にはより簡便な方法が必要である。本研究は、競技者の負担が軽減され、簡便に採取可能な呼気凝縮液中の炎症関連物質の測定方法の開発、および呼吸機能検査との関連性を検討することを目的とした。本研究では、まず、健常者における運動前後の呼気凝縮液を採取して、検討した。(方法)健康な成人男性10名を対象とし、8分間の自転車ペダリング運動前後の呼気凝縮液を採取した。呼気凝縮液中の炎症関連物質としては、cysteinyl Leukotrienes(cysLTs:LTC4、LTD4、LTE4)を測定した。また、運動前後にはスパイロメーターを用いた呼吸機能検査も実施した。(結果)採取した呼気凝縮液中のcysLTs濃度の運動前後における有意な変化は検出できなかった。小児期の気管支喘息の既往がある対象者2名では、運動後に呼気凝縮液量、呼気量および1秒率の低下が観察された。(考察)先行研究では気管支喘息患者で発作時の呼気凝縮液のcysLTsの変化が報告されている。今後、気管支喘息の既往もある競技者や運動誘発性気管支れん縮の競技者において検討する予定である。

  • 高齢者の唾液中ストレスマーカーに関する研究

    【緒言・目的】唾液に含まれる分泌型免疫グロブリンA (SIgA)は、感染の初期防御機構において重要な役割を果たすが加齢により低下すると考えられている。しかし、適度な身体活動によって唾液SIgA分泌が高まることも示されている。そこで本研究では、高齢者の日常における身体活動が唾液SIgA分泌に及ぼす影響について、都市部の対象者で検討すること(課題1)、さらに日常の生活習慣が高齢者の唾液SIgA分泌に及ぼす影響について検討すること(課題2)を目的とした。【方法】課題1の対象は東京都三鷹市在住の前期高齢者118名、後期高齢者166名とし、それぞれ身体活動量を昇順並べて3群に分け(前期高齢者:115および250 kcal/dayを境に3群化;後期高齢者:85および155 kcal/day)、唾液SIgA分泌速度と身体活動量との関係を検討した。課題2の対象は三鷹市在住の前期高齢者23名、後期高齢者15名とした。Quality of lifeを評価するSF-36を用いて各高齢者の身体的健康度と精神的健康度を評価した。それぞれの健康度について、日本国民の平均値と比べて高い群および低い群に分け、SIgA分泌速度との関係を検討した。【結果】課題1:統計的に有意ではないが、前期高齢者では身体活動量が最も多い群、後期高齢者では中程度の群でSIgA分泌速度が他の二群と比較して高い傾向が認められた。課題2:前期高齢者では精神的健康度が高い群のSIgA分泌速度が低い群に比べて有意に高かった(p < 0.05)。また統計的に有意ではないが、後期高齢者では身体的健康度が低い群と比較して高い群においてSIgA分泌速度が高い傾向を示した。また精神的健康度おいても同様の結果が得られた。【考察】課題1において、後期高齢者では1日の身体活動量を85~155kcal/day程度に維持することで、口腔内免疫機能を向上させる可能性が示唆された。課題2では精神的因子が唾液SIgAの分泌に影響を及ぼしている可能性が考えられた。

  • 分泌型免疫グロブリンA産生の加齢変化に対する運動の影響

    (目的)加齢に伴って免疫機能は低下するが、免疫機能は栄養や運動などの生活習慣の影響をうけるため、高齢者において適切な運動は免疫機能を維持向上させる可能性がある。唾液中の分泌型免疫グロブリンA(secretory immunoglobulin A :SIgA)は、経口的に侵入してくる病原微生物に対する一次的防御機構で主要な役割を担う物質であり、唾液分泌速度と唾液SIgA濃度、および両者の積であるSIgA分泌速度は加齢によって低下する。高齢者の免疫機能にとって適切な運動処方について知見を得るために、高齢者のSIgA産生と身体活動量との関係を検討した。(方法)東京都三鷹市の前期高齢者67名と後期高齢者124名を対象として、唾液採取は市販の唾液採取器具(サリベット)を用い、1分間咀嚼させて採取した量を1分間の唾液分泌速度とし、唾液SIgA濃度はELISAで測定した。身体活動量はライフコーダを2週間装着してもらい、その平均値を1日あたりの身体活動量とした。以前測定した前期高齢者283名と後期高齢者119名の身体活動量と今回測定した対象者の身体活動量に有意差が認められなかったため、対象者を合算して解析した。(結果)前期高齢者においては、身体活動量が115kcal/日未満の群に比較して身体活動量が115~250kcal/日の群が有意に高いSIgA分泌速度を示した。身体活動量が250kcal/日以上の群は、身体活動量が115~250kcal/日の群と比較してSIgA分泌速度に有意差はなかった。後期高齢者においては、身体活動量が85~155kcal/日の群でSIgA分泌速度がもっとも高い値を示したものの、85kcal/日未満の群と155kcal/日以上の群とに比較して有意差は見られなかった。(結論)前期高齢者では身体活動量が115 kcal/日以上であるとSIgA産生が多く、後期高齢者では身体活動量以外の因子がSIgA産生に影響を及ぼしていると考えられる。

  • 分泌型免疫グロブリンAの産生に対する加齢と運動の影響

    【目的】加齢に伴う免疫機能低下は運動習慣によってある程度抑制できる可能性がある。唾液中の分泌型免疫グロブリンA(SIgA)を指標として加齢による免疫機能低下に対する運動の効果をヒト(実験1)とラット(実験2)で検討した。【方法】(実験1)後期高齢者32名(78.8±3.1歳)と前期高齢者14名(70.9±2.8歳)の唾液SIgA測定とライフコーダーによる身体活動量測定を行った。(実験2)6ヶ月齢の雄性 SD 系ラット5 匹を飼育して10 ヶ月齢時から1ヶ月ごとに唾液を採取した。コントロールとして8週齢のラット 7 匹の唾液も採取し、SIgA 濃度を ELISAで測定した。【結果】(実験1)後期高齢者ではSIgA分泌速度が39.1±26.3μg/min、身体活動量が100.2±92.8kcal/day、前期高齢者ではSIgA分泌速度が33.6±15.2μg/min、身体活動量が141.2±67.8kcal/dayであった。前年の測定とあわせて対象数を増やして解析すると、前期高齢者ではSIgA分泌速度と身体活動量に正の相関があった。(実験2)10~12ヶ月齢ラットの唾液SIgA濃度は、4.4±2.0(10ヶ月齢)、5.6±1.9(11ヶ月齢)、5.3±2.9(12ヶ月齢)μg/mlで、有意な変動はなかった。8週齢ラットの唾液SIgA濃度は6.6±1.6μg/mlで、10~12ヶ月齢ラットと有意な差はなかった。【考察】ヒトでは、身体活動量が多いとSIgA分泌速度が高い傾向が考えられるが、さらに対象数を増やして検討する必要がある。ラットでは、10~12ヶ月齢では明らかなSIgAの低下は認めなかったが、さらに高齢となる18ヶ月以降に SIgA 濃度の測定、およびSIgA の分泌過程で働く唾液腺内のpolymeric immunoglobulin receptorのmRNA発現を測定し、運動トレーニングの効果も検討する予定で飼育を継続している。

  • 高齢者の上気道感染症防御因子に対する運動の影響

    【目的】加齢にともない免疫機能は低下する.高齢者が健康で活力ある生活を過ごすためには免疫機能の維持が重要である.長期間継続的に行なう適度な運動は,高齢者の免疫機能を改善する可能性があるが,まだ知見が少ない.本研究では,前期高齢者(65歳以上75歳未満)と後期高齢者(75歳以上)において免疫機能と身体活動量との関係を横断的に検討した.【方法】高齢者99名(前期高齢者36名,後期高齢者63名)を対象とした.免疫機能の指標として、1分間に採取した唾液量(唾液分泌速度),唾液中の分泌型免疫グロブリンA(SIgA)濃度,およびSIgA濃度と唾液分泌速度とSIgA濃度との積(SIgA分泌速度)を測定した.身体活動量としては,日常生活による身体活動の消費カロリーをライフコーダー(スズケン社製)によって2週間測定し,その平均値を1日あたりの身体活動量とした.【結果】唾液分泌速度は,前期高齢者において身体活動量と有意な正の相関(p<0.05)が見られたが,後期高齢者では両者に相関が無かった.SIgA濃度は,前期高齢者と後期高齢者ともに身体活動量と相関は無かった.SIgA分泌速度は,前期高齢者において身体活動量と有意な正の相関(p<0.05)が見られた.しかし,後期高齢者では両者に相関がなかった.【考察】前期高齢者では,身体活動量が多いほど唾液分泌速度とSIgA分泌速度が大きく,免疫機能が高いことが示された.しかし、後期高齢者では唾液分泌速度,SIgA濃度,およびSIgA分泌速度と身体活動量との関係はみられなかった.高齢者では加齢による免疫機能低下がおこるので、前期高齢者までに身体活動量を増やすことによって,免疫機能の低下が抑制される可能性も考えられる.
