    Protein kinase C (PKC), upon activation, translocates from the cytosol to the plasma membrane. Phorbol 12-myristate 13-acetate (PMA), a potent PKC activator, is known to induce irreversible translocation of PKC to the plasma membrane, in contrast to the reversible translocation resulting from physiological stimuli and subsequent rapid return to the cytosol (reverse translocation). However, we have previously shown that tyrosine phosphatase (PTPase) inhibitors induce reverse translocation of PMA-stimulated PKCbetaII in porcine polymorphonuclear leukocytes (PMNs). In the present study, we showed that pervanadate, a potent PTPase inhibitor, also induces tyrosine phosphorylation of PMA-stimulated PKCbetaII in porcine PMNs. Furthermore, PP2, a specific inhibitor of Src-family tyrosine kinases (PTKs), was found to inhibit both pervanadate-induced reverse translocation and tyrosine phosphorylation of PMA-stimulated PKCbetaII, suggesting that these two pervanadate-induced responses are mediated by Src-family PTKs. Our findings provide novel insight into the relationship between the subcellular localization and tyrosine phosphorylation of PKC. (C) 2003 Elsevier Inc. All rights reserved.

  • 血清に存在する細胞死誘導物質の単離・同定

    血清に存在する細胞死誘導因子を牛血清から主としてカラムクロマトグラフィーを用いて単離した。活性の検定は、ヒト正常二倍体繊維芽細胞の６～10時間後の細胞致死率の計測によって行った。ブルーセファロース非吸着画分をＤＥＡＥ陰イオン交換カラムで分離し、その後、分子量排除、逆相のＨＰＬＣで分離・生成した。精製化合物を日立Ｍ―4100型高分解能質量分析計により分子量測定を行った結果、分子量181が主成分でその他に分子量476の物質も含まれていた。現在、このいづれが本因子であるかの検討と、他の分析機器を用いた構造解析を進めている。また本因子にはco-factorの存在が示唆されており、その単離・精製も開始する予定である。

  • 培養細胞の血清によるアポトーシスの機構解明

    　本研究で使用した血清は非熱処理の牛胎児血清、細胞はヒト正常二倍体繊維芽細胞を用いた。検定は通常のMEN合成培地に血清の低分子画分を補なった系で行った。これまでの研究で、血清中に存在する血清アルブミンが細胞死誘導活性を有することは解明していたが、それだけでは説明できない。アルブミン以外の細胞死誘導物質の存在が示唆されていた。そこで本研究ではこの未知物質を単離する目的で以下の実験を行った。まず、血清を限外濾過膜（MW cut 1，000）に通し、低分子画分を除去した。次にブルーセファロースカラムに通し、その非吸着画分を得た。この段階で血清アルブミンの混入はELISA測定の感度以下となり、アルブミン以外の細胞死誘導活性の大部分が回収できた。この画分による細胞死はチオールによって救済された。そこで本画分を多量に調製し、それを出発原料として様々なカラムクロマトグラフィーを行った。その結果、DEAEの陰イオン交換クロマトグラフィーが第一段の分離に適していることが分かった。この分離では、非吸着と吸着に大きく分けられたが、本活性は吸着部分に属していた。活性のあるピークを回集し脱塩濃縮後、凍結乾燥し、次に逆相のHPLCで分離を試みた。単一のピークに活性が認められ単離が完了したと思われる。現在、分子量測定、種々の物性計測を行っているところである。

  • 培養細胞の血清によるアポトーシスの機構解明

    本研究の背景は以下に述べる通りである。細胞を培養する際，一般に血清を10％程度添加するのは常識となっている。これは，血清中の増殖因子の供給が主な理由であると理解されている。しかしながら血管内皮細胞や血液細胞を除くほとんど全ての細胞は血清に直接接してはいない。にもかゝわらず培養系に血清を添加するのは，同一ロットが比較的安価に入手できるからである。筆者らは培養系で血清の濃度を増やすと，50％以上で細胞の増殖が悪くなり，60％を越えると細胞死を引き起こすことを見出した。この細胞死はアポトーシスであり，血清の由来や細胞の種類に係りなく起こる。例外は，当然のことながら血管内皮細胞や血液細胞であった。更にこの細胞死はチオール分子によって救済された。 以上のことから本研究では，まず，細胞死を引き起こす本体の解明を試みた。その結果以下のことが明らかになった。 1）血清アルブミンが細胞死を誘導する要因の一つであること。しかしながら，これには血清の低分子画分の存在が必須であり，この本体は数種類のアミノ酸であった。 2）脳背髄液中には血清と同濃度のアルブミンが存在し，しかも同液は細胞死を誘導しないことから，血清中には別の因子の存在が考えられた。現在その因子の単離精製を進めており，分子量1万以上のタンパクである可能性が高い。 3）チオールにより細胞死が救済されることはすでに分かっているが，血清中には更に高分子で細胞死を救済する因子の存在が認められた。これについても現在単離精製を進めている。 今後は上記2），3）の因子の解明に加えて，本現象の細胞内伝達機序の解明を目指していくつもりである。
